如圖所示，SPR 分子相互作用儀器的原理是波長 632.8 nm 的激光形成的 p 偏振光，到達棱鏡后激發金屬表面的表面等離子體發生共振。通過改變入射光角度 θ，實時檢測反射光強度的變化 R，根據反射光強度變化 ΔR 或共振角度的變化 Δθ，實現分子相互作用的檢測；也可以通過固定角度，測量反射光強的變化曲線獲得分子相互作用動力學常數。
